The pMOD4-RT-G plasmid consists of the RT cassette flanked by 50 nucleotide regions identical to the 5' and 3' ends of FLAG-GFP (green) while pMOD4-galK-G consists of the galK cassette flanked by the same sequences. pMOD4-galK-GT consists of the galK cassette flanked by both the FLAG-GFP homology regions and 50 nucleotide regions identical to the 5' and 3' ends of N-terminal and C-terminal TAP (blue and orange, respectively). Both plasmids utilize the R6K-based pMOD4 backbone which is unable to replicate in SW106.
