PCR primers for the genomic region of interest (red box) are designed with attB4 and attB1 sites (blue boxes), and the region is amplified and cloned using a BP reaction into an entry clone. A LR reaction is performed to recombine the entry clone (containing the genomic DNA fragment) into a Tol2 destination vector, placing the genomic fragment upstream of a basal promoter and EGFP (green box).
