Validation of the Mpl antibody. (A) Comparison of two antibodies recognizing Mpl on cryosections from developing Mpl-/- and wild-type mice one day after birth. Left panel: rat monoclonal anti-mouse Mpl (Kirin Brewery), right panel: murine monoclonal anti-human Mpl (R&D Systems). Specific labelling was observed in the brain tissue from wild-type mice (left columns), but was absent in Mpl-/- mice (right columns). (B) Combined in situ hybridization (top row) and immunolabelling (bottom row) on the same sections show that in wild-type mice (left column) the anti-human Mpl antibody (R&D Systems) recognizes the same cells which are labelled by the riboprobe directed against mouse Mpl mRNA. Arrows indicate double-labelled cells.
